The gold standard in quantitative metabolomics
Metabolomics is a powerful tool to answer the multidimensional questions of biology. From just a few microliters of sample, the phenotype of an individual can be uncovered and explored. Metabolomics is key to generating high-quality and reliable data that is mandatory for today’s biological research and individualized health.

The gold standard in targeted metabolomics

Our technology
The central theme of biocrates’ metabolomics profiling products is providing quantitative, reproducible assays designed in an easy and ready-to-use kit format. Quantitative, standardized and quality-controlled methods combined with a standardized format guarantee robust and reproducible results – the cornerstones of any scientific finding.
Biocrates employs a targeted, quantitative approach to all our kits and services. Metabolite panels covering a wide range of analytes are selected based on their application to various metabolic-related diseases and fields of study.
Our assays employ a mix of isotope-labeled internal standards, external calibration curves and quality control samples to generate quantitative results for every analyte. The quantitative nature allows results to be directly compared with the scientific literature or other studies. Having absolute quantification allows for the calculation of the relationships between different metabolites such as sums and ratios of important metabolic indicators.
The ability to generate quantitative data and the standardized format of our kits and services produce results that are reproducible across multiple studies and labs around the globe. The kits, in particular, have been vetted through several ring trials and other longitudinal studies, demonstrating the ability to achieve comparable results from any lab.
All of biocrates’ assays are quality controlled via plasma-based reference samples included with each kit. The accuracy, reproducibility, and quality of results can be quickly and automatically validated within our workflow software. The quality controls also serve as a point of reference for normalization, allowing a greater degree of precision across larger studies spanning long periods of time.
The biocrates kits have been designed with ease of use for the end-user. The kits include all of the required standards and quality controls while utilizing a standardized 96-well plate format. The LC-MS instrument methods and parameters are provided along with the kits, eliminating the laborious method development process. Our software integration simplifies and automates the entire workflow.
Standardized workflow
biocrates’ kits include years of experience in targeted metabolomics. These ready-to-use kits deliver quality-controlled and reproducible quantification of metabolites in biological samples by following only five steps. WebIDQ, a user-friendly workflow manager software, guides the user through the whole process, from sample registration to biological interpretation of the results.

What is in a kit?
The plate
biocrates’ kits are designed for high throughput experiments in a standard 96 well plate format allowing up to 82 user samples per plate. To accommodate users with smaller sample sizes, half sized and training plates are also available. The plates incorporate a novel insert design allowing all preparation and cleanup to be performed directly on the plate. A top filter plate is incorporated to remove proteins that precipitate during preparation. Our proprietary design allows for more efficient drying and extraction improving the quality of results. At the end of preparation, samples are collected in the bottom 96-well plate which can be run directly in your LC-MS autosampler.

Components
biocrates kits are equipped with all of the required components for a robust, quantitative analysis. For quantification, seven levels of calibration standards and isotope-labeled internal standards are used. For performance validation and data normalization, plasma-based quality control samples at three concentration levels are provided and directly incorporated with our software. All reagents come as lyophilized solids in color-coded vials for quick resuspension and easy preparation.

Methods
Each of our kits has been developed and validated for specific LC-MS systems from a number of vendors. We carefully tailor and optimize each method to provide the best possible performance for every instrument. The kits include all vendor-specific acquisition methods. This includes MRM transitions, ion source parameters and chromatography settings to make setup quick and easy without the need for extensive method development. After data collection, processing methods are also provided to make data processing quick and robust. Data processing and validation are directly incorporated with biocrates’ MetIDQ software.
Matrices and sample preparation
While specifically validated for plasma and serum samples, our kits can be used for any samples containing endogenous metabolites. We have developed substantial experience over the years in sample preparation and matrix use.
We offer guidelines on the preparation of blood samples as well as application notes on the preparation of fecal or urine samples and many more. Contact our support in case you have questions on sample preparation or non-standard matrices.
