<p>Although they can be measured with similar analytical methods, lipidomics and metabolomics datasets are often analyzed differently when it comes to statistics. These differences are primarily justified by the biology behind the molecules. In this talk, I will discuss specifically how the nature and structure of standardized lipidomic datasets influences data interpretation and sharing down the line. I’ll introduce tools and approaches to explore lipidomics, and discuss tips and tricks to make the most of this unique type of data. This talk will be especially aimed at scientists who are unsure how to analyze lipidomics and are looking for guidance.</p>

<h2 class="wp-block-heading">Elucidating the impact of food-derived microbial metabolites on intestinal health</h2>

<p>Dietary patterns encompass a diverse array of molecules foreign to the body. Upon ingestion and digestion, these nutrients undergo transformation by the trillions of microorganisms residing in our gastrointestinal tract, which in turn alters the chemical structures of these compounds, affecting their lifespan, bioavailability, and biological effects. Consequently, dietary patterns modulate the gut microbiome, influencing its functions and the production of gut microbial metabolites (GMMs). These GMMs, in turn, play a critical role in regulating gut homeostasis and determining the risk of intestinal disease. To address this complex challenge, we utilize various OMICS approaches. Our research first explores food-gut microbiota interactions by integrating in vitro models, in vivo studies, and omics techniques. We also replicate the gut microenvironment using 3D and 2D intestinal models that accurately represent intestinal homeostasis. Unraveling food-gut microbiome-intestinal interactions promises to pave the way for advancements in personalized nutrition.</p>

<h2 class="wp-block-heading">Standardizing targeted metabolomics studies using Agilent 6495 LC/TQ in concert with biocrates</h2>

<p>Metabolomics and lipidomics are areas of research which have grown hugely over the last 10-15 years. The focus is often on the non-targeted approaches. As knowledge of organism metabolomes and lipidomes has grown, so has the coverage and understanding of metabolic pathways and their networks. Researchers can now focus on key metabolites as markers of well curated pathways known to be impacted by many conditions and changes. This is enabling the emphasis to shift towards targeted approaches. This in turn, provides repeatable sample preparation and analysis with out-of-the-box solutions, such as the biocrates kit. With the new Agilent LC/TQ technologies, the number of metabolites which can be analysed concurrently continues to improve with shorter dwell times, faster polarity switching and increased sensitivity with the aid of improved signal: noise ratios. We show how the Agilent 6495 LC/TQ utilizes these attributes to be a leading multi omics platform for targeted metabolomics, lipidomics and proteomics. <h2 class="wp-block-heading">Data interpretation tools to unleash the full impact of metabolomics</h2>

<p>Data interpretation is the translation of statistically significant results into meaning; from values to pathways and mechanisms, from scattered facts to a structured story. For over 20 years, biocrates has provided its customers with the means to generate high quality metabolomic data, but interpretation remains difficult to standardize. Since I joined the company, I have explored many ways to share my knowledge on data interpretation. First, by providing data interpretation as a service to biocrates customers, next by sharing insights on LinkedIn and in my podcast, The Metabolomist. Last year, I published a guide to the biological interpretation of metabolomics called “The STORY principle”. In this talk, I will discuss the highs and lows of data interpretation for metabolomists, and share the latest tools provided by biocrates to unleash the full impact of metabolomics for research and health.</p>

<h2 class="wp-block-heading">Linking your research question to relevant metabolite features in untargeted metabolomics profiles</h2>

<p>Metabolomics has often been coined as the ultimate phenotyping tool. Indeed, a full understanding of chemical profiles of complex biological extracts would lead to a deeper biochemical understanding of many biological processes. Technological advances made analytical chemical analysis more sensitive than ever before; however, as a result, prioritising the relevant metabolite features that link to your research question becomes increasingly difficult.<br>In this presentation, I will highlight recent advances in computational metabolomics made by my group and others that use networking and machine learning strategies to overcome this challenge. I will introduce FERMO that links phenotypic read-outs to metabolomics profiles in an automatic and reproducible manner to allow for bioactivity-based prediction. Furthermore, I will introduce msFeaST, that combines statistical analyses with feature sets constructed based on machine learning-based embeddings. As both these approaches make use of the experimental design and available metadata thereof, they allow the user to link their research question with the data during the processing and analysis steps. Hence, untargeted metabolomics data analysis will become easier as more time can be spent on the relevant metabolite features.<br>I will finish off with a call to action: sharing is caring! I expect that the presented methodological developments will advance our understanding of the role of metabolites and their complex molecular interactions that underpin growth, development, and health.</p>

<p>Metabolites are the easiest message to quantify in microbiome-host communication. Standardized metabolomics provides a precise functional readout of the impact of microbial communities, anchoring the interpretation of gene-derived functions to the actual effectors that are metabolites. Metabolomics also measures host metabolites that are modulated in response to microbial communities. Thus, the use of standardized and quantitative metabolomics provides the functional readout that bolsters our understanding of the microbiome&#8217;s impact in health and disease.</p>
